Abstract

The commodity chemical succinic acid (succinate) is currently produced frompetrochemicals. Succinate formed through microbial fermentation sequesters carbon dioxide andcould reduce dependence on fossil fuels. This work aims to understand and prolong a high rate ofsuccinate production in a dual-phase fermentation using a genetically modified Escherichia coli.Osmotic conditions resulting from high ionic strength influenced succinate production. Withoutany added osmoprotectant, 53 g succinate accumulated with a yield of 0.78 g/g and aproductivity of 1.29 g/Lh. With 5mM betaine added to the medium succinate accumulated to82.5 g with a 1.01 g/g yield and productivity of 2.01 g/Lh.
