Abstract
Ovate Family Proteins (OFPs) interact with the M8 domain of TONNEAU1-RECRUITING MOTIF family (TRMs) to control organ shape in tomato and other plant species. In this thesis, I sought to investigate the role of OVATE and SlOFP20 (SOV1) as well as various TRMs during tomato shoot regeneration. This transition requires the activation of genes to coordinate cell division and are hypothesized to be regulated by certain OFPs and/or TRMs. I showed that shoot regeneration in Sltrm5 and Sltrm3/4/Sltrm5 were both significantly lower compared to wildtype. An RNA-seq experiment and analysis identified differentially expressed genes in callus and at two shoot regeneration stages; six OFPs (including SlOFP20) and 11 TRMs containing the M8 domain (including SlTRM5) were differentially expressed. This suggests that the gene expression dynamics of certain OFPs and TRMs play a role in cell organization/division at important developmental time points.
